Comparative transcriptome analysis of Trichoderma reesei reveals different gene regulatory networks induced by synthetic mixtures of glucose and β-disaccharide

Abstract The mixture of glucose and β-disaccharide (MGD) synthesized by transglycosylation as a low-cost soluble carbon source can efficiently induce cellulase production in Trichoderma reesei , which holds potential for the biorefining lignocellulosic biomass. However, it is not yet fully understood how MGD induces T. cellulase. In this study, transcriptomic analyses were conducted to investigate molecular basis lignocellulose-degrading enzyme Rut C30 compared with that on lactose. Particular attention was paid CAZymes, transcription factors, transporters other protein processing pathways related lignocellulose degradation. As result, elicit GH5-, GH6- GH7-encoding cellulases up 1.4-fold higher than induced lactose, but GH11- GH74-encoding xylanases are downregulated 1.7- 4.4-fold, respectively. Gene expression profiles suggest activators xyr1 vib1 significantly upregulated mitogen-activated kinase pathway strengthened case lactose induction. addition, hac1- encoding UPR-specific factors MGD, may be enhanced due proper folding nascent proteins. These findings provide theoretical further understanding characterization efficient using an inducer offer strategies strain improvement.
